Abstract

Nonribosomal peptides are a diverse group of compounds synthesized via modular nonribosomal peptide synthetase (NRPS) enzyme complexes. They are produced by a variety of prokaryotes, including streptomycetes, bacillus, and cyanobacteria, and lower-order eukaryotes such as fungi and sponges. NRPS modules often have the ability to incorporate nonproteinogenic amino acids into the synthesized natural product, while structural complexity is achieved by various NRPS tailoring and

modi cation domains. Therefore, a myriad of different peptides can be synthesized ribosomally, far more than those synthesized ribosomally. As such, these bioactive peptides have a varied range of attributes, including antibiotic, antiproliferative, immunomodulatory, metal-sequestering, and toxic properties. In this chapter, we describe the modular nature of NRPSs and the role each domain plays in nonribosomal peptide synthesis. The diverse range of nonribosomal peptide structures and activities are discussed in detail as well as the current eld of combinatorial biosynthesis for creating novel bioactive peptides from these nonribosomal platforms.
