Abstract

[Extract] Although the use of traditional phenol chloroform extraction methods has recently been reported for scabs and cell culture material,94 commercially available extraction kits are currently mainly used to obtain nucleic acids from diagnostic samples. DNA was extracted from tissue samples using the DNeasy Blood and Tissue Kit (QIAGEN, USA). After complete lysis of the specimens by ATL buffer and proteinase K, absolute ethanol was added, then the mixture was transferred to a spin column according to the manufacturer’s protocol.97 The Qiagen AllPrep DNA/RNA extraction kit (Qiagen, Hilden, Germany) was used to extract viral DNA by Lamien and coworkers.25Homogenates of infected tissues (10% weight/volume in PBS), or infected cell culture supernatants were mixed with the kit lysis buffer, RLT plus (200 μL sample + 800 μL buffer). The DNA was extracted according to the manufacturer’s instruc-tions and eluted in 50 μL of elution buffer included in the kit.
